How Much Culture Medium Should Be Added to The Petri Dishes?

Here we talk about the relationship between the size of culture dishes(plates) and the amount of culture medium added.

Many experiments show that the ideal depth of culture medium should be 2-3mm and if the depth is more than 3 mm,It will affect the gas exchange between cells and the outside world and will also cause pollution during transportation.

Based on the above experimental results, we can calculate the common adding amount for a petri dish or culture plate.
Here we take the 6 well culture plate as an example:
The bottom diameter of 6 well culture plate is 3.5 cm,so the radius is 1.75 cm,so we can get the bottom area of each well is:
Π*1.75*1.75=9.62 cm2,so the amount of each well should be 9.62*0.25(here we calculate as 2.5mm depth)=2.4 ml,For convenient measurement, we can count as 2.5ml,if each well is used,we need total 2.5ml*6 wells=15 ml culture medium.
