How to Use Durham Glass Test Tubes for Fermentation Test?
1.Measure a certain amount of culture solution and put it into the test tube (a big test tube).
2.Use a medical syringe to suck a certain amount of culture solution, empty the air, extend the needle to the bottom of the Durham test tube (to prevent bubbles), and slowly push the culture solution until the liquid level rises.Make the solution full.
3.Then add a bit excessive amount of culture solution to wet the wall of durham test tube. Because the culture solution has a certain viscosity, it can make the wet durham glass test tube slowly slide down to the bottom of the test tube. We can also directly point a little culture solution to play a wetting role on the side of the durham test tube.
4.Tilt the large test tube and slide the durham test tube down slowly along the wall of the large test tube, or gently push it with a straw head, etc. Make the Durham test tube slide down slowly.
5.Finish.If there are bubbles, it means failure and needs to be done again.
P.S:
Fermentation is a common biochemical reaction. Different bacteria have different sugar decomposing ability. Some bacteria can break down sugars and produce acids and gases; However, some bacteria only produce acid and no gas, so bubbles cannot be observed by using durham glass test tubes. Therefore, we should repeatedly test to prove the accuracy of the results.
